PURPOSE:To simplify the operation free from effect of plasma along with a higher sensitivity by using 2,2'-azino-bis( 3-ethylbenzothiazoline-6-sulfonate ) (ABTS). CONSTITUTION:A measuring reagent comprises (1) an acetic acid solution of 1.0-4.0mM ABTS, (2) 0.5-3.0% hydrogen peroxide solution and (3) 0.05-0.2M phosphoric acid buffer liquid (pH6-8) containing 0.4-1.2m MEDTA. These reagents are prepared separately and mixed sequentially when hemoglobin in the plasma is determined. To prepare these reagents: (1) ABTS solution: 50-205mg of ABTS is dissolved into 100ml of 80-100% acetic acid solution. (2) hydrogen peroxide solution: 0.5-3ml of 30% hydrogen peroxide solution is prepared by a refined water to 30ml in total. (3) phosphoric acid buffer liquid containing EDTA (ethylenediamine-4-acetate-2-sodium): 2pts. of 0.05-0.2M phosphoric acid- potassium solution containing 0.4-1.2m MEDTA is mixed with 1-3pts. of 0.05- 0.2M phosphate-2-sodium solution containing EDTA with the same concentration and then, controlled to pH6-8 by a 1N sodium hydroxide solution. |
