A recovery and activation process for microbially produced calf prochymosin.

There is disclosed:
a process for recovering microbially produced bovine prochymosin in a form in which it is capable of being converted to active chymosin which comprises:
(a) solubilizing the microbially produced bovine prochymosin in a denaturing solvent; and (b) renaturing the prochymosin by diluting or removing the denaturing agent in the solvent. Further steps leading to the production of active mature chymosin therefrom are also described, these additional steps comprising;
(c) activating the renatured prochymosin by reducing the pH of the solution containing the renatured prochymosin to produce active pseudochymosin; and
(d) elevating the pH of the solution containing the active pseudochymosin to produce active mature chymosin.
