PURPOSE:To separate and to collect an amino acid formed and accumulated in a system, by adding cellulase, cellobiase, and ferrous ion to the fermentation system containing cellulosic substance, cultivating simultaneously a microorganism capable of producing an amino acid. CONSTITUTION:An aqueous solution containing 1-3wt% cellulosic substance such as wood, straw of rice, pericarp of citrus fruit, etc. and proper amounts of N source, inorganic substance, and other nutritive components required for amino acid fermentation is prepared as a substrate. Cellulase, cellobiase, and a microorganism capable of producing an amino acid precultivated and 80- 400ppm ferrous ion are added to the aqueous solution, and the microorganism is fermented in the presence of them. An amino acid formed and accumulated in the fermented solution is separated and collected by a conventional procedure such as ion exchange resin method, etc. The optimum temperature of fermentation operation is 25-40 deg.C, and the fittest pH is in a range of 4-8. L-Lysine, L- threonine, etc. can be prepared as the amino acid. |
