PURPOSE:To obtain a plasmid vector utilizable for the development of a protein in microorganism, by inserting a specific fragment containing ompF promoter of Gram-negative bacteria into a relax-type plasmid originated from Escherichia coli. CONSTITUTION:A relax-type plasmid originated from Escherichia coli, e.g. pBR322, pBR325, pACYC184, etc., especially pBR322, is prepared beforehand. An Alu I fragment, Alu I -BglII fragment or Alu I -Pst I (partially digested) fragment is inserted into the pBR322 plasmid to obtain a plasmid vector. The insertion can be carried out e.g. by incising the plasmid with a restriction enzyme used in the preparation of the above fragment, and linking and cyclizing the obtained DNA fragment with the above fragment using a DNA ligase. |
