NEW MATERIAL:A plasmid having at least two consecutive fractions containing cDNA corresponding to a vegetable viroid RNA. USE:A vector (a gene vector) to insert an exogenote in a vegetable or vegetable cell. PREPARATION:The objective plasmid can be prepared by carrying out the following procedures in the order. (1) The cloning of HSV cDNA by Okayama and Berg's method: (2) preparation of plasmid inserted with monomer or polymer of HSV cDNA: (3) preparation of plasmid inserted with an Escherichia coli lactose operon promoter at the upstream of the monomer HSV cDNA: (4) preparation of plasmid inserted with consecutive polymer HSV cDNA, and (5) transcription of consecutive polymer HSV cDNA to the consecutive polymer HSV-RNA. |
