<p>PURPOSE:To prepare a nucleotide useful as a synthetic raw material of DNA and PNA, stably, in a short time, by reacting a nucleoside with cyclohexylammonium 0-chlorophenyl-beta-cyanoethylphosphate (a novel compound). CONSTITUTION:The nucleotide of formula III is prepared by reacting a nucleoside of formula I [B is (protected) base such as N-benzoylcytosine, N-benzoyladenine, N-isobutylylguanine, thymine, etc.; R<1> is H or OH protected with a group for protecting hydroxyl group of sugars; R<2> is sugar OH-protecting group such as dimethoxytrityl] with a phosphorylation agent comprising the novel cyclohexylammonium o-chlorophenyl-beta-cyanoethylphosphate of formula II preferably at a molar ratio of 1:1-several moles in the presence of a condensation agent at room temperature. The above novel compound can be synthesized e.g. from o-chlorophenyl phosphorodichloridate.</p> |
