PURPOSE:To prepare coenzyme of reduced type NADPH less expensively, by reacting a mold of a bacterium belonging to the genus Methanomicrobium with NADP<+> in a reaction system containing a formate or hydrogen. CONSTITUTION:Novel starin of Methanomicrobium hiroshimaensis(FERM P-7109) is cultivated under anaerobic conditions, and when formation of a gas is stopped, the mold is collected. It is washed, and suspended in a buffer solution, to prepare a mold suspension. By using the mold suspension, coenzyme of reduced type NADPH is prepared from NADP<+> in a reaction system containing a formate or hydrogen. A supernatant liquid obtained after completion of the reaction is made to flow through a DEAE-cellulose column, and subjected to elution of concentration gradient with salt, so that NADPH is separated and collected. |
