PURPOSE:To obtain a novel complex plasmid useful as a cloning vector of Escherichia coli or Bacillus subtilis, by synthesizing it from a plasmid of Streptococcus faecalis and a vector of Escherichia coli. CONSTITUTION:This complex plasmid has tetracyclin resistant gene range derived from plasmid pAMalpha1 of Streptococcus faecalis, ampicillin resistant gene range derived from vector pACYC177 of Escherichia coli, replicating initiation range derived from plasmid pAMalpha1 and replicating initiation range derived from vector pAYC177. Only one recognition scission site in the whole range to restricted enzyme BalI and HpaI is positioned in the tetracyclin resistant scission site, and further only one recognition scission site in the whole range to each of restricted enzyme BglI, PvuI, and PstI is positioned in the ampicillin resistant gene range.
