PURPOSE:To obtain a reagent which can measure accurately and quickly the XIII-th factor for blood clotting by incorporating the red blood cells obtd. by sensitizing the specific antihuman XIII-th factor antibody for blood clotting obtd. by immunizing a rabbit, etc. with the XIII-th factor for blood clotting extracted from the human placenta in the red blood cells of an animal in said reagent. CONSTITUTION:A rabbit or the like is immunized with the XIII-th factor for blood clotting obtd. by refining the extract extracted from the human placenta by a physiological salt soln. and subjected to fractionation and precipitation and the specific antihuman XIII-th factor antibody for blood clotting is obtd. from the antiserum thereof. Such antibody is sensitized with the red blood cells of a sheep, by which the sensitized blood cells are obtd. Such sensitized blood cells are freeze-dried to prepare a reagent. The reagent is added with a phosphoric acid buffer soln. so as to be suspended therein in the stage of measuring the XIII-th factor in the human blood. Such reagent, a positive control and negative control for the human XIII-th factor, and a phosphoric acid buffer soln. are constituted as a kit. The concn. of the XIII-th factor is determined from the max. dilution at which clotting is admitted in comparison with the negative control. The measurement up to the dilution of 1/10,000 concn. of a normal person is thus made possible. |
