PURPOSE:A solution containing human immune interferon (I-IFN) resultant from gene recombination technique is puified with a monoclonal antibody binding with I-IFN to give substantially pure I-IFN. CONSTITUTION:Cells excreting human I-IFN such as human peripheral lymphocytes are cultured and the mRNA coding I-IFN is separated from the culture mixture, and, for example, a single-chained CDNA is synthesized using a reverse transcriptase and introduced into double-chained DNA and plasmid to cause, e.g., transformation of Escherichia coli, from which the double-chained DNA coding I-IFN is obtained. The host is transformed with the resultant DNA and cultured to form and accumulate I-IFN and the solution resultant from the bacteriolysis is purified by means of a monoclonal antibiody binding with I-IFN to obtain the I-IFN protein of a specific activity over 10<7>U/mg having aminoacid sequence of the formula (Y is Met, binding bond; Z1, Z2 are Cys, 1/2Cys).
