PURPOSE:Plasmid pTP1102 having a large number of copies, tetracyclin resistance and chloramphenicol resistance, having tetracyclin-resistant gene, chloramphenicol-resistant gene, specific molecular weight, and cleavage point map of restricted enzyme. CONSTITUTION:Plasmid pTP1102 having tetracyclin-resistant gene, chloramphenicol-resistant gene, about 4.3 megadalton molecular weight, and a cleavage point map of restricted enzyme shown by the figure. Plasmid pTP10 is separated from Corynebacterium xerosis. Bacillus subtilis is transformed with it, plasmid (pTP11) is reseparated. It and plasmid PNSI derived from Staphylococcus are scissored with restricted enzyme hind III, and ligated, to give plasmid pTP1102 having about 4.3 megadalton molecular weight. |
