PURPOSE:To prevent attraction of optical isomers except the optical isomer intended to be separated and to enable the sepn. of the optical isomer with easy sepn. efficiency in a short time by using the particles formed by immobilizing an optically active amino acid on the pore surface of silica gel as a packing material for a liquid chromatography. CONSTITUTION:A silane coupling agent such as 3-amino-propyl triethoxy silane or the like is brought into reaction with the pore surface of silica gel, thereby forming porous spherical particles of aminopropylated silica gel. An optically active amino acid having a butyl oxycarbonyl group, etc. as a protective group and a reagent (1,1'-carbonyl di-imidazole, etc.) into which an activated N-acyl group can be introduced are brought into reaction with such particles, by which the particles immobilized with the optically active amino acid are obtd. Chromatography is performed by using the silica gel particles as a packing agent, by which the sepn. of an optical isomer of various amino acids, mandelic acid, etc. can be efficiently performed. The packing material has high strength, is easy to handle and contributes to an increase in the immobilizing capacity of the amino acid. |
