PURPOSE:To measure surely a slight amt. of antigen by making use of the phenomenon that the clotting of a carrier sensitized with a complete antigen (referring to antigens except hapten) and a carrier sensitized with a single kind of monoclonal antibody against said antigen is prohibited by the antigen in a specimen (patient's serum, etc.). CONSTITUTION:A single monoclonal antibody is drawn from the cell obtd. by the cell fusion of the antibody forming cell obtd. by a subcutaneous injection of the complete antigen of hormone, alpha-fetoprotein, serum protein, etc. in an animal and a suitable myeloma cell and said antibody sensitized with, for example, polystyrene latex particles is prepd. On the other hand, the complete antigen sensitized with polystyrene latex particles is preliminarily prepd. Each one drop of said antigen-sensitized latex and anti-body sensitized latex is added to one drop of a prescribed dilute liquid of a patient's serum, etc. and both are uniformly mixed and after 2min, the presence or absence of the clotting image is observed. If there is an antigen in the serum, etc. or up to a certain dilution rate, the clotting image is not observed. The detection and determination of the slight antigen are thus made possible with good reproducibility in a short time. |
