PURPOSE:To separate a plasmid utilizable as a carrier of genetic information from microorganisms belonging to Pediococcus genus. CONSTITUTION:A microbial strain having a plasmid and belonging to Pediococcus genus is cultured in a medium, and the microbial cells are collected from the cultured product and subjected to bacteriolysis. The objective plasmid pSP-2 having a restriction enzyme incision map of the figure is separated from the bacteriolysis product. Any microorganisms can be used for the preparation of the plasmid provided that it belongs to Pediococcus genus and contains the plasmid. The bacteriolysis of the collected cells can be carried out by any methods capable of lysing the cell and extracting the plasmid DNA in a perfect form, preferably by a method using e.g. a lysing enzyme such as lysozyme. |
