PURPOSE:To obtain a cell line for preparing hybridoma derived by mutation from tumor cells other than a tumor cell derived from marrow cell. CONSTITUTION:A cancerous cell is irradiated with ultraviolet rays or provided with a mutagen such as ethylmethanesulfonate, N-methyl-N'-nitro-N-nitrosoguanidine, etc. in a proper dose or amount depending upon its kind, the cell is transferred to a medium containing 8-azaguanidine, to develop a strain resistant to 8-azaguanidine. The use of HAT selection is then examined. The cell is cultivated, the medium is removed by centrifugation, the cell is washed with a HAT medium, and transferred to the same HAT medium. It is observed for several days, and life or death is judged. |
