摘要 NEW MATERIAL:The hexacosapeptide of formula. USE:An agent to inhibit fibrinolysis. It has high affinity to plasmin. PREPARATION:Human plasma is treated with a plasminogen.sepharose 4B column to obtain alpha2-PI having a molecular weight of about 67,000 (abbreviated as 67Kalpha2PI), which is converted to two kinds of proteins, i.e. the one having a molecular weight of about 65,000 (65Kalpha2PI) and having a molecular weight of about 60,000 (60Kalpha2PI) by SDS-polyacrylamide gel electrophoresis. The product is subjected to gel-filtration and separated into a high-molecular fraction and a low-molecular fraction. The high-molecular fraction is subjected to ultrafiltration and then treated with a plasminogen-sepharose 4B column to separate the 67Kalpha2PI from 65Kalpha2PI and 60Kalpha2PI. The difference of the structures of the eluted fractions was compared according to a trypsin map, and the structure fragment existing only in the 67Kalpha2PI was treated with high performance liquid chromatography. The separated fraction was subjected to amino acid analysis. The peptide was determined to have the structure of the formula.
