PURPOSE:To efficiently form spores, by adding an alkali to a medium containing a specific nutritive source and keeping the medium containing a specific nutritive source in a specific pH range in a spore formation period. CONSTITUTION:A starch and a seasoned solution, meat extract, peptone, yeast extract, etc., as a nitrogen source are added to the culture tank T, sterilized, cooled to 80-90 deg.C, incorporated with a culture solution containing spores of Clostridium butyricum, cooled to 37 deg.C and adjusted to 7.0pH. The adjustment of pH is carried out with 10wt% NaOH or KOH. The mixture is allowed to stand at 7.0pH for 2-3hr, the spores are made into nutritive cells, multiplication is started, the mixture is kept for 3-5hr while adjusting it to 7pH. Then, the pH is gradually lowered. When the advance of multiplication reaches the maximum, the mixture is kept at 6.0pH for about 1hr, the multiplication is sufficiently advanced, the mixture is adjusted to 6.0-5.5pH and kept for 3-6hr. |
