PURPOSE:To obtain a plasmid having the desired marker in high frequency, by preparing a conjugated plasmid having an easily separable marker in order to select a recombined DNA. CONSTITUTION:A DNA fragment obtained by scissoring a plasmid having a genetic range to become a marker or a chromosome DNA fragment with a restricted enzyme for recognizing 6 base and a DNA fragment of cryptic plasmid obtained by scissoring a cryptic plasmid having no marker with a restricted enzyme recognizing 4 base, producing a scission end having complementarity with a scission end of the restricted enzyme for recognizing 6 base are prepared. These DNA fragments are linked to give a plasmid, and a conjugated plasmid containing a genetic range directing properties of introduced marker and drive unit of cryptic plasmid, multiplying in a host microorganism and developing the properties of marker is selected with the plasmid. |
