NEW MATERIAL:The manifestation vector containing the recognition sequence of restriction enzyme having a promoter, a ribosome-bonding site and a translation initiation signal, and remaining the 3'-protruded terminal connected to the translation initiation signal as incised form. USE:A manifestation vector to integrate a DNA fragment to code polypeptide, and develop said polypeptide in high efficiency. PREPARATION:A manifestation vector having a promoter, a ribosome-bonding site and a translation initiation signal, and containing a recognition sequence of a restriction enzyme remaining a 3'-protruded terminal as incised form following the translation initiation signal, is incised with a restriction enzyme, and the produced 3'-protruded terminal is converted to a flat terminal with DNA polymerase I or T4DNA polymerase. An exogenic gene is integrated in said falt terminal, and the hostmicroorganism is transformed by the obtained recombinant. The resultant transformed strain is cultured to manifest the exogenic gene. |
