PURPOSE:To determine ATP in high sensitivity, by forming reduced NAD from ATP at a high molar ratio. CONSTITUTION:ATP is separated from a kinase, ADP and a phosphorus compound for kinase substrate. The obtained solution or a specimen liquid containing ATP is made to react with a fatty acid and if necessary with CoASH to form acyl-CoA, which is converted successively to dehydroacyl-CoA, hydroxyacyl-CoA, ketoacyl-CoA and acyl-CoA. The amount of reduced NAD formed in each cycle of acyl-CoA is determined by measuring the absorption at the specific absorption wavelength, or measuring the color development of the hydrogen- transmission chromogen having hydrogen-accepting capability. |
