Highly active pancreatin is obtained by autolysis of an aqueous isopropanol-containing tissue homogenate which is buffered with, preferably, sodium bicarbonate until a test precipitation with aqueous isopropyl alcohol has a positive result and precipitation with increasing concentration of isopropyl alcohol, resulting in a suspension of fibres which can be removed by screening and permits pancreatin to be isolated directly from the solution by further increasing the concentration of isopropyl alcohol.
