A plasmid for a periplasmic or extracellular bacterial protein was cleaved; a double-stranded DNA sequence coding for a selected protein or protion thereof from a eukaryotic cell, such as insulin, was inserted in that cleaved gene by recombinant DNA technique and used to transform a bacterium, and the excreted selected protein was collected. Thus, double stranded cDNA prepd. from RNA contg. preproinsulin in RNA isolated from a B-cell tumor was inserted into an Escherichia coli penicillinase gene.
