A compound suitable for use in the determination of a terminal amino acid of a polypeptide possesses the general formula D-C-B-A wherein A is a moiety which can react with and bind to a terminal amino acid of a polypeptide molecule and can result in the removal of the terminal amino acid; B is a moiety which provides steric separation or other desirable properties between C and A; C is a moiety that can be detected directly or which can be derivatized with a group that can be detected; and D is a moiety which protects the C moiety from degradation or other modification during reaction with the terminal amino acid.
