NEW MATERIAL:The plasminogen.activator having the following characteristics: exhibits single protein band by the electrophoresis using sodium dodecylsulfate/ polyacrylamide gel; apparent molecular weight, 70,000+ or -5,000; the pI of the main band of isoelectric point electrophoresis is 7-9; immunological property characterized by the lack of adsorptivity by the affinity chromatography using antiurokinase IgG-Sepharose; exhibits hydrolytic activity to the substances of table I and does not exhibit the activity to the substances of table II. USE:Thrombolytic agent. PROCESS:The human kidney is cleaned by removing coagulated blood, connective tissue, lipid, etc. therefrom, and extracted with a buffer solution, or with 1-2M NH4SCN (7.4pH) by perfusion. The extract liquid is passed through plural columns such as a column packed with an ion exchange material, a column containing metal chelate, etc. to obtain purified plasminogen.activator. The activator can be adsorbed essentially completely to a fibrin Sepharose column, and eluted by 2M NH4SCN solution.
