<p>Isosorbide 5-nitrate of formula (I) is made by (1) reacting isosorbide with p-nitro benzoyl chloride in organic solvent such as tetrahydrofurane or methylene chloride and in presence of pyridine or triethylamine, to obtain isosorbide 2-nitro benzoate of formula (II); (2) reacting the prod. with nitric acid in acetic acid, acetic anhydride or mixt. of these, to obtain 5-nitro isosorbide 2-p-nitro benzoate of formula (III); and (3) reacting the prod. with sodium or potassium hydroxide to give (I).</p> |
