PURPOSE:To determine an enzyme accurately in a short time, by liberating a specific compound, reacting the liberated specific compound with an oxidase in the presence of a coupler, and measuring the amount of sensible change. CONSTITUTION:A hydrolase is reacted with a synthetic substrate containing a compound of the formula[R1 is OH group, amino or H when one or more of R2-R6 are OH group or amino group ; R2-R6 are H, halogen, lower alkyl, lower alkoxyl, (substituted) amino, OH group, carboxyl or sulfo or R5 and R6 together may form a ring] as a constituent factor to liberate the compound of the formula, which is then reacted with an oxidase, e.g. ascorbic acid oxidase, capable of consuming at least oxygen and developing a color with a coupler, e.g. salicyclic acid. The resultant developed color is then determined with light of 500-750nm wavelength by the colorimetry. |
