PURPOSE:To carry out the quick measurement of uric acid concentration of blood, by determining uric acid using an enzymatic reaction by immobilized uricase obtained by treating a carrier prepared from a PVC-based resin solution with a uricase solution. CONSTITUTION:A vinyl chloride-based resin is dissovled in a solvent (solvent A), good solvent for the resin, processed into a liquid film having a given shape, and brought into contact with a solvent (solvent B) which is a poor solvent for the resin and a good solvent for the solvent A, to give a carrier. A solution containing >=50mg/dl uricase in a buffer solution at 8-9pH is prepared, and the carrier is immersed in tis solution, to give immobilized uricase. Uricase converts uric acid into allantoin in the presence of an enzyme. To determine uric acid using the immobilized uricase, for example, a cell is equipped with an oxygen sensor wherein an immobilized uricase film is attached to an oxygen electrode or hydrogen peroxide electrode, a specimen is introduced into the cell, and the amount of oxygen consumed by the reaction or the amount of hydrogen peroxide evolved is measured, to determine uric acid. |
