<p>PURPOSE:To prepare plasmid pNS1981 useful as a plasmid vector of Bacillus subtilis, by the DNA infection of a specific DNA fraction of tetracycline-resistant plasmid pNS1 originated from staphylococcus to Bacillus subtilis devoid of plasmid. CONSTITUTION:A specific DNA fraction of a tetracycline-resistant plasmid pNS1 originated from staphylococcus and capable of being replicated stably in Bacillus subtilis (e.g. the A-fraction of Hind III decomposition product), is introduced by DNA infection into the protoplast prepared from the cultured cells of a bacterial strain belonging to Bacillus subtilis, e.g. Bacillus subtilis Marburg GSY908 (FERM-P No. 6469), and the obtained bacterial strain containing pNS1981 is cultured and the objective plasmid pNS1981 is separated therefrom. pNS1981 has a molecular weight of about 3.1 mega-dalton, exhibits resistance to tetracycline, and is incised with restriction enzyme at the points shown in the acission point map.</p> |
