PURPOSE:To analyze the peptide in vital body fluid, etc. with high sensitivity, by separating a peptide component with a high velocity charomatography from a peptide-contg. sample, adding an alkali soln. to the eluate and hydrolyzing the same under heating then cooling the soln. and detecting the fluorescence by an o-phthalaldehyde reagent. CONSTITUTION:A peptide-contg. sample in a peptide analyzer 1 is supplied from a sample introducer 5 to a high velocity chromatographic column 6, and a moving phase 3 (contg. 0.02mol potassium dihydrogenphosphate) is fed to separate a peptide component. A 1-2 normal soln. of NaOH or the like is fed to the eluate from a storage tank 8 and while the soln is passed through the inside of a reaction tube 11 which is a stainless steel tube heated to around 160 deg.C, the peptide component is hydrolyzed. The separated liquid is cooled to room temp. with a cooling tube, whereafter an o-phthalaldehyde reagent is fed from a tank 15 and the intensity of the generated fluorescence is measured with a detector 17. Thus, the long chain peptide which could not be measured with the prior art is made analyzable with good sensitivity. |
