PURPOSE:To purify the titled antibiotic in high yield, by adsorbing an aqueous solution containing neothramycin A and/or neothramycin B on a strongly basic anion exchanger, followed by subjecting it to chromatography for eluting it with an aqueous solution of a neutral salt. CONSTITUTION:A culture filtrate containing neothramycin A wherein R<1> is OH and R<2> is H in a compound shown by the formula and nethramycin B wherein R<1> is H and R<2> is OH is treated with a nonionic porous resin (e.g., divinylbenzene-styrene copolymer) so that neothramycin is adsorbed on it. Neothramycin is eluted with 5-50wt% acetone-containing aqueous solution or diluted solution of NH3 in water, to give a partially purified aqueous solution, which is directly adsorbed on a strongly basic anion exchanger (e.g., polystyrene resin having a quaternary ammonium group as an active group) or lyophilized and redissolved in the solution to give an aqueous solution, and the aqueous solution is adsorbed on the anion exchanger. Neothramycin is eluted with an aqueous solution of a neutral salt (e.g., NaCl), and treated with a nonionic porous resin. These operations are repeated so that neothramycin is purified. |
