<p>PURPOSE:The cultivation of a bacterium producing EF-70147-Y, an antibiotic, in Streptomyces is conducted aerobically in an enriched culture medium to enable high-efficiency isolation and collection of EF-70147-Y from the culture mixture. CONSTITUTION:Carbon sources such as glucose, glycerol, nitrogen sources such as soybean flour or meat extract and inorganic salts such as calcium carbonate or sodium chloride are used for the culture medium for Streptomyces sp 7014, when necessary, aminoacids and vitamins are further added. The cultivation is effected by the aerobic submerged process at 18-35 deg.C for about 24-240hr. EF-70147-Y, accumulated mainly in the liquid part of the culture mixture, is subjected to an appropriate combination of adsorption and desorption with an ion-exchange resin, gel-filtration chromatography and extraction with a variety of organic solvents to effect isolation and purification.</p> |
