L-Tryptophan was manufd. by tryptophan synthetase and tryptophanase from the reaction of indole with L-serine. Thus, E. coli MT-10232 was inoculated into a medium contg. beef extract 1, peptone 0.5, yeast extract 0.1, and KH2PO4 0.2% (pH=7.0), and incubated at 30≦̸C for 20 hrs. with shaking, to obtain tryptophan synthetase. P. putida IFO-12996 was inocu-lated into the medium contg. beef ext. 1.0, peptone 1.0, and NaCl 0.5%, to obtain serine racemase. Triton X-100 contg. indole, DL-serine, Na2SO3, and pyridoxalphosphate were mixed and reacted with trp. synthetase and ser. racemase to give 28.5g L-trp.
