Creation of DNA sequences encoding modified proinsulin precursors.

Methods and compositions are provided for the production of insulin. DNA sequences encoding modified proinsulin precursors are produced in vitro and inserted into production cells wherein modified proinsulin precursors are elaborated. The modified proinsulin precursors are cleaved in vitro to yield insulin. Modified proinsulin precursors comprise aspartic acid at the amino terminus of the C chain and/or methiorine at the carboxyl terminus of the C chain.
