PREPARATION OF HEAT-RESISTANT PROTEASE

摘要

PURPOSE:To prepare a heat-resistant protease, in high productivity, by cloning the structural gene of heat-resistant protease derived from a thermophilic bacterium using a thermophilic bacterium as a host, and amplifying the gene. CONSTITUTION:The protease structure gene of a heat-resistant protease (e.g. Bacillus stearothermophilus ATCC12980) originated from a thermophilic bacterium is integrated into a vector plasmid pTB90 (resistant to kanamycin and tetracycline) using a thermophilic bacterium as a host by shot-gun method. After the amplification of the gene, it is transformed by protoplast process to obtain the objective heat-resistant protease.
