The immobilisation of glucose isomerase is enhanced by first impregnating the matrix with at least 0.1 mmole Mg(2+) per g of matrix. Pref. amt. of Mg(2+) is 0.1-2 mmole/g. Impregnation is pref. by contact with a soln. of (in)organic Mg salt, pref. the halide, sulphate, nitrate, hypophosphite, fluorosilicate, acetate or lactate. The soln. is pref. 1-25 mM in Mg(2+). MgSO4 is the pref. salt. - Pref. the support is an inorganic oxide impregnated with a polyamine which is then crosslinked with excess of a bifunctional reagent to provide pendant functional gps. - The half-life of the immobilised enzyme is increased, and time for the immobilisation may be decreased. The enzyme may be used for converting glucose to fructose. DEAB- DE3224024 C Glucose isomerase is immobilised on a carrier matrix by treating the matrix with at least 0.1 mmoles of divalent Mg ions, pref. 0.1-2m moles MgSO4 per Gram carrier matrix before contact with the enzyme.
|
