PURPOSE:To enable the rapid and accurate determination of choline esterase, by using a choline esterase determination reagent containing acetic acid kinase and adenosine triphosphate. CONSTITUTION:When acetylcholine is used as a substrate and made to react with the choline esterase (ChE) in the biospecimen, the substrate is decomposed to acetic acid and choline. The acetic acid is converted to acetyl phosphate using adenosine triphosphate as a cosubstrate when the system contains acetic acid kinase. The acetic acid kinase has substrate specificity only to acetic acid. The ChE can be determined by the determination of the produced acetyl phosphate or adenosine diphosphate. The determination of acetyl phosphate can be carried out by the colorimetry at the visible region after reacting with a proper chromogen, and adenosine diphosphate can be determined by using a conjugated enzyme system of pyruvic acid kinase and lactic acid dehydrogenase.
