A technique for purification and concentration of urokinase is described. Urokinase was adsorbed to the affinity column at pH>=6 and eluted from the column at pH<=4.5. Thus, a urokinase soln with a concn.5, 200 IU/M1 and a purity of 370 IU/mg protein was layered on an aprotinin-EACA-Sepharose column preequilibrated with 1M-NaCl and 0.02M phosphate buffer, pH 7.2. The column was washed with the buffer soln. to remove protein impurities and then washed with 1M-NaCl. Absorbed urokinase was eluted from the column with 1M-NaCl, pH 3.9. The yield of urokinase was 71.5% with a purity of 61,000 IU/mg protein.
