PURPOSE:To prepare the titled enzyme usable as a clinical testing reagent for the specific determination of lecithin in a phospholipid, by separating and collecting the titled enzyme from a culture of a microorganism, belonging to the genus Streptomyces, and capable of producing the titled enzyme. CONSTITUTION:A microorganism, e.g. bacterium AA 586 strain (FERM-No.6100), belonging to the genus Streptomyces, and capable of producing phospholipase D-P is cultivated by the conventional method for producing enzymes. The cultivation is usually carried out by the liquid cultivation method, and advantageously carried out by the submerged culture method with aeration for industrial purpose. The cultivation is completed at 25-30 deg.C for 40-90hr. The resultant enzyme is readily soluble in water and capable of producing the aimed phospholipase D-P by the well-known method. The enzyme is a novel enzyme, exhibiting a high substrate specificity for lecithin in phospholipids, ad having the phospholipase D activity of catalyzing a reaction of the lecithin to form phosphatidic acid and choline. |
