PURPOSE:To determine discriminatively d-nicotine and l-nicotine in a mixture of d-nicone and l-nicotine in a short time in high sensitivity and in high accuracy, by using an immobilized bacterium sensor. CONSTITUTION:The total amounts of d-nicotine and l-nicotine are determined by a bacterium sensor obtained by immobilizing a bacterium such as Arthrobactor globiformis IFO12956, etc. capable of producing dl-nicotine oxidase, capable of measuring electrochemically dissolved oxygen which is prepared by the bacterium through the assimilation of nicotine depending upon the concentration of nicotine, and simultaneously the amount l-nicotine is selectively determined by a bacterium sensor obtained by immobilizing a bacterium such as Peseudomonas putida JTS9, etc. capable of producing l-nicotine oxidase. The amount of d-nicotine is obtained from the difference of the two. |
