<p>Method for the enzymatic replacement of a C-terminal amino acid in an insulin or insulin-like compound with another amino compound or for the addition of an amino compound to a des-insulin or des-insulin-like compound. In the preferred embodiment the method permits the conversion of swine insulin to human insulin in high yield and in short reaction times by preparing a reaction mixture comprising an insulin or insulin-like compound having a first C-terminal amino acid; an amino compound or salt thereof; a solvent system comprising a buffer, capable of maintaining the pH of the reaction mixture between about 5 and 7, and between about 75% and 97% (vol/vol) of at least one non-aqueous reaction mixture miscible solvent, said reaction mixture miscible solvent including at least about 50% (vol/vol) butane-1, 4-diol, and between about 3% and 25% (vol/vol) water; and at least one protease specific to the cleavage of said first C-terminal amino acid from the insulin or insulin-like compound and at least one protease capable joining to that insulin or insulin-like compound said amino compound or salt thereof, but substantially incapable of cleaving the insulin or insulin-like compound elsewhere under the conditions used; and maintaining the temperature of the mixture between about 4 DEG C and 6 DEG C for about 1 to 2 hours.</p> |
