发明名称 QUANTITATIVE DETERMINATION OF ENDOTOXIN
摘要 PURPOSE:To measure the concentration of endotoxin in high sensitivity by using a small quantity of an extracted component of a red blood corpuscle of a horseshoe crab, by reacting the extracted component of the red blood corpuscle of the horseshoe crab to a solution containing the endotoxin in a plastic chassis and measuring the necessary gelation time. CONSTITUTION:0.02ml extracted component of a red blood corpuscle of a horseshoe crab (called ''Richet solution'', available in market) is dropped in 0.02ml sample water (city water, distilled water, desalted water etc.) sampled in a chassis and said chassis is covered and the gelation time after reaction is observed by the naked eye rocking lightly the chassis at five minutes intervals in 37 deg.C thermostat. A curve of the gelation time is found by a dilution system of preliminarily known quantity of endotoxin (unnecessary to find a standard curve at every time of measurement) and the concentration of the endotoxin is known by said curve. 0.05mg/l endotoxin is measured in high sensitivity by using about one tenth conventional Richet solution.
