PURPOSE:A DNA which hybridizes DNA sequence of N-4ACMCase gene and codes polypeptide having cellulase activity, obtained by a transforming host from a chromosome DNA fragment of Bacillus sp. No.N-4 strain. CONSTITUTION:Chromosome DNA of Bacillus sp. ATCC21833 strain is scissored with restriction enzyme Hind III, linked to vector pBR322 similarly scissored with the enzyme in the presence of ligase and transduced to Escherichia coli to give plasmid pNK2 containing Hind III fragment with 2.1kb and plasmid pNK2 containing Hind III fragment with 2.8kb from a transformant. Plasmid pNK1 to code cellulase is approximately equal to plasmid pNK2 except a fact that the plasmid pNK1 has 60 direct repeat amino acid sequence in the vicinity of C-terminal end. Cellulase obtained from the plasmids substantially no difference from cellulase produced by N-4 strain. |
