NEW MATERIAL:Relax type cyclic plasmid pMS76 having scission sites against various kinds of restriction enzymes consisting of one site by BamHI, one site by SamI, and one site by PstI, unscissored by EcoRI, SalIor Hind III, having a DNA site of ampicillin resistance at a distance within 5.6Kb-8.48Kb- 1.3Kb, from the scission site of BamHI and about 5.54 megadalton molecular weight. USE:The novel plasmid pMS76 has properties like those of Col EI plasmid such as amplification by chloramphenicol, compatibility with another plasmid, etc., useful as one of vectors to be used in genetic recombination requiring plural different genes, needing respectively different vectors. PROCESS:Separated and collected from a culture of Esherichia coli (K-12ML-5,000 strain (FERM-P 82) having resistance only to ampicillin. |
