RECOMBINANT PLASMID CONTAINING HUMAN BETA-INTERFERON GENE

PURPOSE:To provide a recombinant plasmid capable of transforming Escherichia coli and producing human beta-interferon (beta-IFN) in extremely high efficiency, by integrating a DNA fraction coding beta-IFN to the downstream of the tryptophan promoter of Escherichia coli. CONSTITUTION:A bacterial strain prepared by the lysogenization of transduction phage (lambdaptrp) carrying tryptophan operon in Escherichia coli to induce the phage, purifying the obtained phage lytic solution to obtain lambdaptrpDNA. The DNA is integrated in plasmid pBR325 to obtain a plasmid vector pKYP-1 containing trp promoter. The fraction containing trp promoter and SD sequence obtained therefrom is integrated in pBR322 to obtain pKYP-5, which is made portable and inserted successively with two trp promoters to obtain pKYP-12. The objective recombinant plasmid pLE-3 can be obtained by integrating beta-IFN- coding DNA in the pKYP-12.
