PURPOSE:To make it possible to anylize guipenosides in Hydrangea Dulcis Folium ext. while completely separating them into individual components, by employing a reversed phase partition filler and an ultraviolet detector of wavelength 200-210nm. CONSTITUTION:Hydrangea Dulcis Folium ext. is dissolved in water, and guipenosides are extracted on shaking by means of water-saturated n-butanol. After the solvent is removed from the extracts, the residue is dissolved in methanol to prepare a sample, which is injected to a liquid chromatograph. Then, with a reversed phase partition filler and a mixture of water and a water miscible organic solvent defined as a stationary phase and a mobile phase, respectively, the flow rate of the mobile phase is stepwisely raised to separate the guipenosides into individual components. Then, each of the separated components are detected by means of an ultraviolet detector of wavelength 200-210nm, and the peak height of the obtained chromatogram is compared with that of a reference substance, thereby obtaining the amount of each guipenoside contained in the sample. |
