PURPOSE:A hybrid plasmid useful for developing blended protein of human corticotropin-beta-lipotropin (ACTH-beta-LPH), obtained by using plasmid pA22 as a vector. CONSTITUTION:A DNA fragment derived from a gene of ACTH-beta-LPH precursor is introduced to the EcoRI site of pA22, cyclic plasmid having at least one resistant gene, self-replicating function, and EcoRI site of resticted enzyme just behind an amppromotor operation region and initiation codon ATG, obtained from plasmid such as pBR322, pBR325, etc. having two or more resistant genes containing amplicillin resistant gene. The prepared hybrid plasmid is introduced into a host such as E. coil HB 101, x1776, etc. and blended protein ACTH-beta-LPH is developed. |
