PURPOSE:A hybrid plasmid useful for developing blended protein of human corticotropin-beta-lipotropin(ACTH-beta-LPH), obtained by using plasmid pYG1 as a vector. CONSTITUTION:A DNA fragment derived from a gene of ACTH-beta-LPH precursor is introduced to the EcoRI site of plasmid pYG1, having at least one resistant gene, self-replicating function, and the DNA fragment comprising a lacpromotor operator region and the first several amino acid codons of beta-galctosidase between the site of restricted enzyme EcoRI and the site of restricted enzyme AvaI or the site of a restricted enzyme having only one clevage part in tetracycline resistant gene. pYG1 is obtained by digesting pBR322 with EcoRI and AvaI to separate a fragment, which is linked to the DNA fragment derived from phage M13mp2. |
