PURPOSE:A plasmid useful for developing genetic information, having a specific DNA fragment between the site of restricted enzyme EcoRI and the site of restricted enzyme AvaI or the side of a restricted enzyme having only one cleavage part in tetracycline gene. CONSTITUTION:A plasmid such as pBR322, pBR325, etc. having the site of EcoRI and the site of AvaI or the site of a restricted enzyme with the only one cleavage part in tetracycline resistant gene, having one or more resistant genes such as ampicillin, etc. after the digestion by them, and self-replicating function, is digested with EcoRI and AvaI, and separated by electrophoresis, to give a long fragment. While phage vector M13mp2 is degested with EcoRI and AvaI to give a DNA fragment comprising a lacpromotor operator region and the first five amino acid codons of beta-galactosidase, and both the fragments are bonded to give the desired plasmid pYG1. |
