PURPOSE:To recover streptolysin-O in high concentration in high purity, by using the isoelectric point of streptolysin-O adsorbed on a carrier having anion exchange ability. CONSTITUTION:The culture filtrate of a micrococcus of hemolytic chain Streptococcus pyogenes is brought into contact with a column of carrier having anion exchange ability such as diethylaminoethylated crosslinked agarose at the isoelectric point of streptolysin, namely at a pH value higher than pH 5.2- 5.4, so that streptolysin-O is adsorbed on the carrier. A buffer solution having a pH value lower the isoelectric point is introduced to the column, a gradient ranging from a pH value higher than the isoelectric point to one lower than the isoelectric point is produced in the column, the buffer solution is continuously fed to the column, the part having the pH value lower than the isoelectric point in the column is extended to the outlet of the column, so that streptolysin-O is concentrated in the column, and it is taken out separately. |
