PURPOSE:To analyze unstable polymer protein which is eluted under strong acidity without denaturation of protein by neutralizing said protein quickly. CONSTITUTION:The sample loaded by a sample injector 2 is separated and eluted of protein in a reverse phase column 3 by the solvent fed from a liquid feed pump 1. When the intended protein in the sample is detected with a detector 4, the valve of a changer 5 for the flow passage of sampling is rotated by 60 deg. to the left, by which the eluate is held in loops A' B. Upon completion of the holding, the valve of the changer 5 is rotated by 60 deg. to the right to return the loop A' to the state of A. The loop B is connected to a liquid feed pump 11, and the acidic org. solvent loaded and held directly in a column 13 for GPC is neutralized by the aq. solvent delivered from the pump 11 and the org. solvent is removed in the column 13. The dissolved stable protein is separated and eluted and is analyzed by a detector 14 for GPC. |
